Skin is one of the organs highly susceptible to radiation. It is important to investigate the influence of radiation exposure on the skin at the molecular level. Recently, we have identified the genes regulated by radiation exposure in hairless mice, using indirect comparative microarray analysis. In the present study, we investigated the gene expression profile of x-irradiated hairless mouse using direct comparative microarray. After microarray analysis, we identified 50 genes that showed up-regulation at 13 day after x-ray irradiation. These include stefin A3, S100A8 and loricrin, similar to the data obtained by indirect microarray analysis. The expression of selected genes was further verified by RT-PCR, and showed highly up-regulated gene expression by x-ray. Our results demonstrate the complexity of the transcriptional profile of the radiation response, providing important clues on which to base further investigations of the molecular events underlying radiationinduced skin injury.
